Criteria for Shedder Status
As described in Johannessen et al. (2021), doi: 10.1016/j.fsigen.2021.102541: DNA depositions after firmly holding a sterile plastic tube for 30s; categories of high, medium and low shedders according to the DNA strength (total RFU value) and the quality of the profile.
Previous Activities
wash both hands with soap and water for 15 seconds and let their hands air-dry, then attend 1 hour seminar with one hand (active) normal activities and one hand (inactive) not touching anything
Contact Scenario
holding plastic tube for 30s
Primary Substrate
Primary Substrate Type
plastic tube
Primary Substrate Material
Deposit
holding for 30s, direct swabbing of 2x2cm2 squares of forehead
Delay

Sampling
Background DNA on Sampled Surface
Sampling Time
sampling within 1 hour after deposition
Persistence
< 1 hour
Sampling Method
Tubes: cotton swab (Selefa) moistened with 60 µL 0.9% NaCl, sweeping the upper tip of the cotton head in parallel movements while the swab was rotated slowly;
Sampling Area
tubes: entire surface, except for cap
Laboratory Analysis
Extraction

DNA Quantification
RB1 qPCR assay on a LightCycler Nano instrument (Roche); specific volumes and reagents specified in chapter 2.4 "DNA quantification and STR analysis"
Input for Profiling
N/A
Profiling
PowerPlex ESX 16 Fast System (Promega Corporation), HID Veriti Thermal Cycler, ABI 3500 Genetic Analyzer (36 cm capillary array; POP4; injection parameters: 13 s/1.2 kV; analytical thresholds: blue channel 30 rfu, green 45 rfu, black (yellow) 70 rfu and red 75 rfu)
Reference Samples
yes, buccal swabs from all participants
Profile Interpretation and Mixture Analysis
≤ 3 foreign alleles counted as drop-in, foreign alleles in stutter position of donor allele removed if peak height <20% of donor allele,
RNA Data Interpretation
N/A
Results
DNA Quantity
active hand: 0.06-9.7; inactive hand: 0-3.9ng
Profile Quality
mostly complete profiles for active hand with foreign alleles present in 49% of samples; more incomplete profiles for inactive hand with foreign alleles present in 37% of samples (supplement)
Parameter Used for Comparison
number of alleles detected, percentage donor vs foreign alleles, percentage of donor vs foreign dna (TPH), total DNA amounts, statistical tests (ANOVA)
Summary of Results
strong correlation of DNA amount on hands and face indicate that source of touch DNA is not (only) from hands but from other body parts that are touched/rubbed/scatched
Raised Questions
more knowledge behind DNA shedding status required. How does it behave under certain conditions?
Cautionary Remarks
N/A
