Chelex: significantly better from cotton than from plastic, DNA IQ: minimally better from cotton than from plastic (mostly not significant); Plastic: DNA IQ performs better than Chelex, Cotton: both extraction methods perform more or less equally well; DNA yield variation higher when manual Chelex extraction method used; conclusion: extraction efficiency according to substrate type needs to be taken into account when comparing DNA yields
