Results

DNA Quantity

avg. 495 pg from fired and unfired Wolf brand caliber olive steel cartridges

Profile Quality

strong partial profiles or better using MiniFiler amplification, no profiles using Powerplex BIO

Parameter Used for Comparison

DNA yield (ng), profiles quality (full, strong partial (>50% expected alleles), weak partial, no profile (no alleles at all))

Summary of Results

DNA IQ extraction methods are not significantly different, whereas the organic extraction method yields significantly lower amounts of DNA; the DNA yield from fired cartridges is not significantly lower than from unfired cartridges; the average profile completeness is not significantly lower from fired than from unfired cartridges (roughly 80-85% of expected alleles) using MiniFiler; No profiles obtained using PowerPlex 16 Bio kit or Identifiler kit; No consistency observed in developing full profiles among ammunition type;

Cautionary Remarks

determination method of shedder status n.s.; exact handling scenarios not always clear from methods section (e.g. subsequent handling by different participants in study testing various ammunition types not described in methods section)
